The dimerization of deoxyhemoglobin and of oxyhemoglobin. Evidence for cleavage along the same plane.

چکیده

Studies on mixtures of canine and human hemoglobin incubated in the liganded form and separated in the unliganded form (and vice versa) indicate that liganded and unliganded hemoglobin cleave along the same plane in dimer formation. The separation was carried out by isoelectric focusing in polyacrylamide gels. The same result was indicated by experiments in which the canine-human hemoglobin mixture was focused in the oxy form and then refocused in the deoxy form as the oxygen in the gel reacted with dithionite. This technique clearly illustrates that, as a result of the Bohr effect, liganded and unliganded hemoglobin have dhferent isoelectric points. Electrofocusmg in gels could profitably be used to study cooperativity in allosteric enzymes.

G-protein Coupled Receptor Dimerization

A growing body of evidence suggests that GPCRs exist and function as dimers or higher oligomers. The evidence for GPCR dimerization comes from biochemical, biophysical and functional studies. In addition, researchers have shown the occurrence of heterodimerization between different members of the GPCR family. Two receptors can interact with each other to make a dimer through their extracellular...

Comparison of oxyhemoglobin and deoxyhemoglobin signal reliability with and without global mean removal for digit manipulation motor tasks.

Functional near-infrared spectroscopy (fNIRS) could be well suited for clinical use, such as measuring neural activity before and after treatment; however, reliability and specificity of fNIRS signals must be ensured so that differences can be attributed to the intervention. This study compared the test-retest and longitudinal reliability of oxyhemoglobin and deoxyhemoglobin signals before and ...

I-38: Chromosome Instability in The Cleavage Stage Embryo

Recently, we demonstrated chromosome instability (CIN) in human cleavage stage embryogenesis following in vitro fertilization (IVF). CIN not necessarily undermines normal human development (i.e. when remaining normal diploid blastomeres develop the embryo proper), however it can spark a spectrum of conditions, including loss of conception, genetic disease and genetic variation development. To s...

The effect of potassium chloride on the Bohr effect of human hemoglobin.

The normal and differential titration curves of liganded and unliganded hemoglobin were measured at various KCl concentrations (0.1 to 2.0 M). In this range of KCl concentrations, the curves for deoxyhemoglobin showed no salt-induced pK changes of titratable groups. In the same salt concentration range oxyhemoglobin showed a marked change in titration behavior which could only be accounted for ...

Applications of higher order shear deformation theories on stress distribution in a five layer sandwich plate

In this paper, layerwise theory (LT) along with the first, second and third-order shear deformation theories (FSDT, SSDT and TSDT) are used to determine the stress distribution in a simply supported square sandwich plate subjected to a uniformly distributed load. Two functionally graded (FG) face sheets encapsulate an elastomeric core while two epoxy adhesive layers adhere the core to the face ...
